My guess is somebody bought some handstamps at a flee market then had a laugh putting them on the cover.
Also, I can clearly see “Jeff” in the “autograph” so looked up an actual Jefferson signature and it matches nicely. So I suspect someone thought it would be funny to replicate his signature but ran out of steam after the first few letters so just did flourishes.
Haven't considered the date yet.

"My guess is somebody bought some handstamps at a flee market then had a laugh putting them on the cover."
So here's a strange cover I found in a big box of "stuff".
Thomas Jefferson coil stamp of 1968 on Artmaster Cachet.
Then you notice a red star on each stamp, a row of penguins, a "Return To Sender" hand, a couple of "PAID" stamps and some sort of autograph from January 16, 1934. Up under the flap someone had marked 570 $16.00 ...
Can anyone solve this riddle?
